Effect of Cycloporin to Puromycin Aminconucleoside Induced Nephrosis in Rats

Abstract
This experimental study was conducted to determine the effect of T-cell immunity, as judged by changes observed by cyclosporin administrstion, to PAN induced nephrosis in rats. Sprague-Dawley rats weighing 150-200 gm were used and divided into controls, Group I (PAN intraperitoneally) and Group II (PAN intraperitoneally, followed by cyclosporin subcutaenously for 2 weeks). Twenty four-hour urinary protein excretion was measured on day 0, 7, 14 and 2l. On the 21st day, rats were sacrificed for determination of total serum protein, albumin and cholesterol levels. And anionic sites of lamina rara extema was determined by using PEI as cationic probes. The following results were obtained. Twety four-hour urinary protein excretion (mg/day) was significantly increased to 185.2?94.9 on the 7th day compared to 2.9?1.2 on day 0 (p<0.01), and de- creased gradually to 40.9?33.7 on the 21st day. No differences was not noted between Group I and Group II. Total serum protein and albumin did not show significant difference in Group I and II compared with controls. However, serum cholesterol levels were higher than controls. The number of anionic sites per 1 pm length of glomerular basement memberane in Group I and II were 10.4?1,1 and 10.2?1.7 respectively. And these values were significantly lower than control value, 14.6?1.2. However, there was no difference between Group I and II. In light microscopic observation, interstitial infiltration of monuclear cells was noted in Group I and II, with more prominent degree in Group II. In summary, present study, except for inducing mononuclear cell infiltration in renal interstitium, did not demonstrate any significant changes of renal epithelial cells by PAN, suggesting that T-cell immunity had no significant effect on PAN-induced nephrosis in rats.
